You can click boxes and type in fields on that settings page just like you can anything else. The easiest thing to do is to start with recording a test and walking through the steps that you want to take. You may have to adjust that test a bit but that's a good place to start.

It works for Firefox, but I m unable to spy any such element in chrome browser

As per https://support.smartbear.com/testcomplete/docs/app-testing/web/general/considerations.html#Chrome :
"Chrome refuses all extensions to inject code into its internal pages (about:*, chrome:*). Because of that, internal pages, like the new tab page, the extension manager, the download manager, the history page and so on, are not accessible to TestComplete."

I'd agree, The best starting point is always to hit record and see how TC picks up the objects.
If it has trouble picking up every action, you may be able to create the test steps manually.

How can we set downloads path for firefox from testcomplete so that I can verify for a downloaded file in a particular path
