Precise answer to your questions requires more details from you, but in a general, I think that you should be able to test mainframe application and likewise your web services if they conform to the current restrictions.

You should be able to test mainframe application using the approach for console applications (http://support.smartbear.com/viewarticle/29636/). As for web services testing and requirements see http://support.smartbear.com/viewarticle/26970/.

If for some reason (mostly - because of used security) TestComplete appears to be not able to send the proper requests to web services, SoapUI (Pro) seems to be not a bad choice as well.
