I've been unable to replicate the issue you are describing, unfortunately.
Ok here is a work around
If I clone the data source then the new or the cloned one has a the new coulmn
its a difficult one to repro.
One way to do it is create and shut down READYAPI and then add a column on startup
When this problem occurs, are there any indications of something failing in the ReadyAPI logs?
